Any seasoned blues musician will tell you that the simplest and most effective way to improve is by creating the perfect practice environment and practice, practice and practice some more.
Check out the links to these two articles below. I have always been big into shred guitar, but I have found out over the years the blues truly are the root of most styles, and that you absolutely MUST be able to grasp the FEEL of the blues, and integrate that feel and groove into any style of music you play, be it rock, country, soul whatever, you MUST practice in a blues style and environment for a period of time at least long enough to have blues influence your style.
Even if your goal is not to be a complete blues player, once you learn how to get the feel of playing with blues musicians, it will give your playing a whole different feel, and believe me, that feel will be noticed by you and your listeners. You will also bring a whole new feel to you personally when you sit down just you and you guitar and enjoy each others company.

Best thing you can do is listen to the masters. Over and over again. Go to the web (myspace, youtube, iTunes, etc) …
Find the bluesmen and women. Listen to their work. Figure out riffs, make new ones … etc.
